Separation of alphavirus nucleocapsids from envelope fragments.

Methods for the preparation of alphavirus nucleocapsids and envelope fragments for biochemical, electron microscopical and serological investigations are described. Difficulties which have been encountered previously in attempts to isolate viral nucleocapsids for morphological studies, namely disruption of the fragile particles, have been overcome by treatment with formaldehyde.

Ubiquitin depletion and dominant-negative VPS4 inhibit rhabdovirus budding without affecting alphavirus budding.

The budding reactions of a number of enveloped viruses use the cellular machinery involved in the formation of the luminal vesicles of endosomal multivesicular bodies (MVB). Budding of these viruses is dependent on the presence of specific late-domain motifs in membrane-associated viral proteins. Such budding reactions usually involve ubiquitin and are blocked by expression of an ATPase-deficie...

Role of cell surface spikes in alphavirus budding.

Alphaviruses mature by budding at cell surfaces. According to a prevailing hypothesis, the viral membrane protein, which is a heterodimeric protein unit, is transported to the plasma membrane (PM), where it awaits binding to the viral nucleocapsid (NC). This hypothesis predicts that the viral membrane protein heterodimers accumulate at the cell surface when expressed in the absence of NCs. We h...

Lexical Features are Not Necessary for Sequence Labeling

We use the technique of SVM anchoring to demonstrate that lexical features extracted from an annotated training corpus are not necessary to obtain state of the art results on tasks such as Named Entity Recognition and Chunking. While standard models require as many as 100K distinct features, we derive models with as little as 1K features that perform as well. These robust reduced models indicat...

Infectious Agents Are Not Necessary for Murine Atherogenesis

Recent work has revealed correlations between bacterial or viral infections and atherosclerotic disease. One particular bacterium, Chlamydia pneumoniae, has been observed at high frequency in human atherosclerotic lesions, prompting the hypothesis that infectious agents may be necessary for the initiation or progression of atherosclerosis. To determine if responses to gram-negative bacteria are...
